Gene Optimization Service

Production of genetically modified organisms (GMOs) with traits such as increased yields, disease resistance etc., is still largely based on naturally occurring gene sequences. However, use of synthetic genes customized for increased protein expression in exogenous system is a technique followed by many scientists today. Ocimum provides efficient and quality driven service to optimize your genes to suit expression in organism of choice, and also to manipulate sequence for ease in cloning.

Native gene sequences are optimized through incorporation of desirable features using our proprietary software, OptGene™. Any optimization carried out will only incorporate silent mutations, and change the native sequences minimally. Gene sequences will be optimized for following features:
  • Adaptation of host codon usage and utilization
  • Directed mutagenesis, insertion and removal of chosen RE sites
  • Knockout of cryptic splice sites, RNA destabilizing sequences and other undesirable sequence signals
  • Removal of secondary structures in RNA
  • Reduction of transcription regions in unused frames
What You Need to Provide:
  1. Accession numbers or DNA or protein sequence.
    Multiple sequences may be submitted in the same file.
  2. Source organism - Organism from where the sequence was obtained.
    Standard codon tables will be used as default.
  3. Target organism – Organism whose genetic codon and codon usage tables will be used.
    Customized codon and frequency tables may also be provided.
  4. Patterns/motifs to avoid preferentially.
    A 'Preferred or Must' option must be selected, for use in scoring of final optimizations.
  5. RE sites to remove, and again a 'Preferred or Must' tag.
  6. RE sites to insert and which positions (Please provide a range).
  7. Sequences of linkers you need to add at 5' and 3' ends of the optimized gene.
    Please note that these sequences will be used as these are, and not optimized sequences.
What we will send you:
  1. Top three optimized sequences.
  2. Scoring method used. This will be based on client-defined preferences.
  3. A report with step-by-step track of all modifications made to your sequence.
  4. Results may either be downloaded or delivered on a CD.
